ZSYY001 is a novel nanoparticle formulation of polymeric micellar paclitaxel (PM-paclitaxel) designed to improve paclitaxel delivery while avoiding the side effects associated with Cremophor EL, the solvent used in conventional paclitaxel formulations. ZSYY001 utilizes polymeric micelles to solubilize paclitaxel, enabling intravenous administration without the need for premedication. In preclinical studies, ZSYY001 demonstrated significant tumor growth inhibition with minimal toxicity. In a phase I clinical trial in patients with advanced solid tumors, ZSYY001 showed a favorable safety profile, no acute hypersensitivity reactions, and promising antitumor activity, with partial responses and stable disease in several patients, including those previously treated with paclitaxel. Dose escalation up to 390 mg/m^2 did not reach a maximum tolerated dose, and the pharmacokinetic profile was nonlinear.
