Overview

ZT-22 is a synthetic benzofuran derivative of beta-elemene, a bioactive sesquiterpene isolated from the medicinal plant Curcuma wenyujin. It is currently under preclinical investigation as a potential therapeutic agent for hepatocellular carcinoma (HCC). ZT-22 demonstrates significantly enhanced anti-tumor efficacy compared to its parent compound, beta-elemene, in both in vitro and in vivo models. Its mechanism of action involves direct binding to Heat Shock Protein Family A Member 6 (HSPA6), which subsequently triggers the activation of the p38 mitogen-activated protein kinase (MAPK) signaling pathway. This activation leads to the inhibition of HCC cell proliferation and the induction of apoptosis. The binding of ZT-22 to HSPA6 has been validated through various techniques, including drug affinity-responsive target stability (DARTS), cellular thermal shift assay (CETSA), and surface plasmon resonance (SPR).
