ZT003

ZT003 is an investigational engineered dual agonist fusion protein that targets both the glucagon-like peptide 1 receptor (GLP-1R) and the fibroblast growth factor 21 receptor (FGF21R) pathway, being developed primarily for obesity, type 2 diabetes, metabolic dysfunction–associated steatohepatitis (MASH/NASH), and severe hypertriglyceridemia. Preclinical data describe ZT003 as a GLP-1/FGF21 dual agonist fusion comprising a GLP-1 moiety, an anti–human serum albumin nanobody for half-life extension, and an FGF21 component conjugated to a small molecule to prevent C‑terminal degradation, producing synergistic effects on weight loss, glycemic control, lipid metabolism, and liver steatosis with weekly dosing potential.[2][4][6] First‑in‑human randomized, double‑blind, placebo‑controlled phase 1 studies are evaluating single and multiple ascending subcutaneous doses of ZT003 in healthy, overweight, or obese volunteers to characterize its safety, tolerability, pharmacokinetics, and immunogenicity.[1][3][5][12]
