ZTx101 is a first-in-class, gut-restricted small molecule inhibitor developed by Zehna Therapeutics. It targets the gut microbial enzyme trimethylamine (TMA) lyase (specifically the CutC/D complex) to prevent the conversion of dietary choline into TMA. By inhibiting this initial step in the gut, ZTx101 aims to reduce systemic levels of trimethylamine N-oxide (TMAO), a metabolite strongly associated with the pathogenesis of cardiovascular disease (CVD), chronic weight management, and metabolic indications. As a gut-restricted agent, it is designed to act locally within the intestinal lumen with minimal systemic absorption, thereby reducing the risk of off-target effects.
