**ZVX-60** is an allogeneic, off-the-shelf cell-based vaccine developed by Heat Biologics using their proprietary gp96 platform combined with OX40L-Fc and SARS-CoV-2 spike protein. It expresses secreted heat shock protein gp96-Ig as a potent adjuvant that chaperones tumor or viral antigens to activate antigen-presenting cells via Toll-Like Receptor 4/2 (TLR4/TLR2) signaling, specifically inducing HLA-A2 restricted CD8+ T-cells and enhancing CD4+ T-cell responses. OX40L-Fc provides costimulation to boost antigen-specific T-cell expansion, memory formation (effector and central memory CD8+ T-cells), and antibody production (anti-Spike IgG and IgM). Preclinical data in mice showed robust systemic and lung tissue-resident memory T-cell responses at low doses (0.5 million cells), with upregulation of activation markers like CD69 and CXCR6, positioning it for prophylactic use against COVID-19, potentially alone or combined with other vaccines to improve protection in vulnerable populations.[1][3][10]
