ZW1528 is a novel bispecific antibody developed by Zymeworks that targets both interleukin-4 receptor alpha (IL-4Rα) and interleukin-33 (IL-33). It is designed to simultaneously block the signaling pathways of IL-4, IL-13, and IL-33—key mediators in respiratory inflammation. The molecule leverages Zymeworks’ proprietary Azymetric technology to achieve high-affinity binding to both targets, resulting in potent inhibition of Type 2 and non-Type 2 inflammatory responses. Preclinical studies have demonstrated its efficacy in suppressing cytokine-driven activation of human epithelial cells and immune cells from COPD patients, as well as effectiveness in murine models of lung inflammation. The antibody features an IgG-like structure with Fc modifications for extended half-life and stability at high concentrations, supporting subcutaneous administration. ZW1528 represents the company’s first development candidate for autoimmune and inflammatory diseases such as chronic obstructive pulmonary disease (COPD), with regulatory filings for Phase 1 clinical trials expected in the second half of 2026[1][3][4][7][10].
