Overview

ZW171 is a novel bispecific antibody developed by Zymeworks for the treatment of advanced or metastatic cancers that express mesothelin (MSLN), including ovarian cancer, non-small cell lung cancer (NSCLC), mesothelioma, and other solid tumors. It utilizes a unique "2+1" T-cell engager format, simultaneously binding to the extracellular domain of MSLN on tumor cells and CD3 on T cells. This dual targeting enables potent T cell-mediated tumor cell killing while aiming to minimize on-target, off-tumor toxicity and cytokine release syndrome through incorporation of a low-affinity anti-CD3 binder. The antibody is engineered using proprietary Azymetric and EFECT technologies to enhance selectivity for tumor tissue over normal tissue, improving both safety and efficacy in preclinical models. As of mid-2025, it is being evaluated in global Phase 1 clinical trials for multiple MSLN-expressing cancers[1][2][4][6][7][9].
