ZW27941 is a potent small-molecule degrader of Methyltransferase-like 3 (METTL3), the catalytic subunit of the N6-methyladenosine (m6A) RNA methyltransferase complex. By inducing the degradation of METTL3, ZW27941 disrupts the post-transcriptional regulation of oncogenic programs in Acute Myeloid Leukemia (AML). Preclinical studies in MOLM13 cells demonstrate that ZW27941 treatment leads to the downregulation of key AML drivers, lipid metabolism genes, and transcription factors involved in lineage identity, while simultaneously activating apoptotic pathways and immune signaling (including TNFα, NF-κB, and JAK-STAT3). It is being investigated as a therapeutic strategy to overcome leukemic cell survival and drug resistance by targeting the m6A regulatory machinery.
