ZW327 is a novel antibody-drug conjugate (ADC) developed by Zymeworks for the treatment of cancers that overexpress Lymphocyte antigen 6E (Ly6E). It consists of a highly specific Ly6E-targeting monoclonal antibody conjugated to a proprietary topoisomerase 1 inhibitor payload, identified as ZD06519. The ADC is designed to bind and internalize into Ly6E-expressing tumor cells, delivering the cytotoxic payload directly to malignant cells while sparing normal tissues. Preclinical studies have demonstrated robust anti-tumor activity across various cancer models—including non-small cell lung cancer (NSCLC), triple-negative breast cancer, head and neck squamous cell carcinoma, and gastrointestinal cancers such as pancreatic ductal adenocarcinoma. ZW327 has shown favorable pharmacokinetics and tolerability in non-human primate studies at exposure levels above those projected to be efficacious[1][3][4][5][8].
