Drug intelligence / Profile preview
ZW33 is a biparatopic, bispecific anti‑HER2 antibody‑drug conjugate (ADC) developed by Zymeworks for the treatment of HER2‑expressing solid tumors, including ovarian, breast, and gastric cancers. It is built on the same bispecific antibody framework as ZW25, which binds two distinct epitopes on human epidermal growth factor receptor 2 (HER2/ERBB2), and is conjugated to a cytotoxic payload to selectively deliver chemotherapy to HER2‑positive tumor cells while limiting systemic toxicity.[1][2][7][5] ZW33 has received FDA orphan drug designation for ovarian cancer but has not advanced beyond preclinical development, and the overall research program is listed as having no active clinical development reports as of 2022.[2][4][5]
