ZW418 is a bispecific T-cell engager (BiTE) designed to target B7-H3 (CD276) and CD3. Developed by Zymeworks using their proprietary Azymetric™ and EFECT™ platforms, the molecule is engineered to redirect T-cell cytotoxicity against B7-H3-expressing tumor cells. B7-H3 is an immune checkpoint protein frequently overexpressed in a wide range of solid tumors, including lung, prostate, and breast cancers, and is associated with poor prognosis and immune evasion. ZW418 incorporates specific Fc modifications to reduce the risk of systemic cytokine release syndrome (CRS) while maintaining potent anti-tumor efficacy. It is currently being investigated for the treatment of advanced B7-H3-positive solid tumors.
