ZW427 is a novel antibody-drug conjugate (ADC) being developed by Zymeworks for the treatment of RAS-mutated cancers. It targets Ly6E (Lymphocyte Antigen 6 Complex, Locus E), a protein frequently overexpressed in various solid tumors, including non-small cell lung cancer (NSCLC), pancreatic cancer, and colorectal cancer. ZW427 is engineered to deliver a novel pan-RAS inhibitor payload directly to tumor cells, aiming to overcome the efficacy and safety limitations associated with current small-molecule RAS inhibitors. Preclinical studies have shown that ZW427 possesses potent tumor-killing capabilities and significant bystander activity. As of early 2026, the candidate is in preclinical development.
