ZW439 is a novel antibody-drug conjugate (ADC) being developed by Zymeworks for the treatment of pancreatic cancer and other tumors driven by RAS mutations. It consists of an antibody targeting Claudin 18.2 (CLDN18.2) conjugated to a novel pan-RAS inhibitor payload. Preclinical studies have shown that ZW439 exhibits potent and targeted tumor cell killing across various CLDN18.2 expression levels, along with significant bystander activity and a favorable tolerability profile. The drug is designed to address the high unmet medical need in RAS-mutated cancers by delivering the inhibitory payload directly to CLDN18.2-positive cells.
