ZX-8177 is a potent, small-molecule inhibitor of ecto-nucleotide pyrophosphatase/phosphodiesterase-1 (ENPP1) developed by Nanjing Zenshine Pharmaceuticals. ENPP1 is the primary enzyme responsible for the hydrolysis of 2'3'-cyclic GMP-AMP (cGAMP), which is a natural ligand for the Stimulator of Interferon Genes (STING) pathway. By inhibiting ENPP1, ZX-8177 prevents the degradation of cGAMP, thereby enhancing STING activation and promoting an anti-tumor immune response. This mechanism induces the production of interferon-beta (IFN-β1) and facilitates the recruitment of M1 macrophages, natural killer (NK) cells, and CD8+ T cells into the tumor immune microenvironment. ZX-8177 is currently being evaluated in Phase 1 clinical trials for the treatment of advanced unresectable, recurrent, or metastatic solid tumors, both as a monotherapy and with potential for synergistic effects when combined with chemotherapy or radiotherapy.
