Overview

ZXP-8202 is a small molecule inhibitor of ectonucleotide pyrophosphatase/phosphodiesterase 1 (ENPP1) developed by Zenshine Pharmaceuticals. ENPP1 is a membrane-bound nucleotide hydrolase that degrades cyclic GMP-AMP (cGAMP), a key activator of the stimulator of interferon genes (STING) pathway. By blocking ENPP1-mediated hydrolysis of cGAMP, ZXP-8202 prevents the degradation of this immune transmitter, thereby promoting STING-dependent innate immune activation and interferon-beta (IFN-beta) production in the tumor microenvironment. In preclinical studies, ZXP-8202 demonstrated potent enzymatic inhibition and achieved significant tumor growth inhibition in syngeneic mouse models. Development of ZXP-8202 has been discontinued in favor of other pipeline candidates such as ZX-8177.
