ZY19489 is a novel, first-in-class triaminopyrimidine small molecule antimalarial drug developed for the treatment of malaria, including both Plasmodium falciparum and Plasmodium vivax infections. It was discovered through a collaboration between AstraZeneca and Medicines for Malaria Venture (MMV), with development led by Zydus Lifesciences (formerly Cadila Healthcare). The compound demonstrates potent activity against blood-stage parasites, including drug-resistant strains, and has shown rapid parasite clearance in human challenge studies. Its mechanism of action is currently undefined but appears to be novel; no resistance has been detected in vitro or in clinical studies to date. The drug is being investigated as a potential single-dose cure for uncomplicated malaria and may be used in combination with other agents due to limited activity against mature gametocytes and liver-stage parasites[5][6][7][8].
