Drug intelligence / Profile preview
Zyprexa Adhera is a long-acting intramuscular injectable formulation of the atypical antipsychotic olanzapine, developed by Eli Lilly. It consists of the salt olanzapine pamoate monohydrate, which is designed to provide sustained therapeutic levels of olanzapine over a period of two to four weeks. The drug is primarily indicated for the treatment of schizophrenia and has been evaluated for schizoaffective disorder. Its mechanism of action involves potent antagonism at dopamine D2 and serotonin 5-HT2A receptors, which helps manage both positive and negative symptoms of psychosis. Due to the risk of post-injection delirium sedation syndrome (PDSS), a rare but serious condition characterized by sedation and/or delirium following injection, Zyprexa Adhera is administered under a restricted distribution program (REMS in the United States) that requires patients to be monitored for at least three hours post-administration in a healthcare facility.
