Overview

ZZW-115 is a potent small molecule inhibitor of NUPR1 (Nuclear Protein 1), a stress-induced protein implicated in cancer cell survival and tumor progression. It binds to NUPR1 with a dissociation constant (Kd) of 2.1 μM and inhibits its function by blocking the translocation of NUPR1 from the cytoplasm to the nucleus, likely through competition with importins[7][1]. Mechanistically, ZZW-115 induces tumor cell death via necroptosis, apoptosis, and ferroptosis in a mitochondria-dependent manner[3][4][8]. Preclinical studies have demonstrated that treatment with ZZW-115 significantly reduces pancreatic intraepithelial neoplasia (PanINs) and blocks NUPR1-dependent stress granule formation in mouse models of pancreatic ductal adenocarcinoma[6]. The compound also leads to reactive oxygen species (ROS) accumulation followed by ferroptotic cell death. Its primary development focus is as an anticancer agent.
