Overview

β-1,2-mannotriose-containing Candida cell wall epitopes are specialized carbohydrate structures located within the phosphomannan complex of the Candida albicans cell wall (Han & Cutler, 1995). These epitopes are unique to fungi, as the β-1,2-linkage is absent in human glycans, making them highly specific targets for therapeutic intervention (Bundle et al., 2005). Biologically, these mannotriose units facilitate the adherence of the fungus to host tissues and contribute to immune evasion by masking other cell wall components (Xin & Cutler, 2011). In the context of disease, they are primary antigenic determinants during invasive and mucosal candidiasis. Therapeutic strategies targeting these epitopes include the development of monoclonal antibodies, such as MAb B6.1, and synthetic glycoconjugate vaccines designed to elicit protective humoral immunity. By binding to these epitopes, drugs can promote opsonization by phagocytes and neutralize the virulence of the pathogen (Johnson et al., 2012).

Mechanism of action

Targeting these epitopes via monoclonal antibodies or vaccines induces opsonophagocytosis and inhibits the adherence of Candida species to host epithelial and endothelial cells, thereby preventing colonization and systemic dissemination.
