The β-hematin crystal formation pathway is a critical detoxification process employed by Plasmodium parasites during the intraerythrocytic stage of malaria (Egan, T. J., 2008, J. Chem. Biol.). As the parasite digests host hemoglobin within its acidic food vacuole to obtain amino acids, it releases large quantities of free heme (ferriprotoporphyrin IX), which is highly toxic due to its ability to generate reactive oxygen species and disrupt cellular membranes (Hempelmann, E., 2007, Parasitol. Res.). To survive, the parasite converts this toxic heme into an insoluble, chemically inert crystalline form known as hemozoin, of which β-hematin is the synthetic equivalent (Sullivan, D. J., et al., 1996, Science). This biomineralization process is the primary target for several classes of antimalarial drugs, most notably the quinolines. By binding to the growing faces of the crystals or forming complexes with free heme, these drugs prevent sequestration, leading to an accumulation of toxic heme that ultimately kills the parasite (Pagola, S., et al., 2000, Nature). Understanding this pathway is essential for developing new treatments to overcome widespread drug resistance mediated by parasite transporter mutations.
