Overview

The γ-aminobutyric acid A receptor is a pentameric ligand-gated chloride ion channel located primarily on neurons in the central nervous system. It mediates the inhibitory effects of GABA, the principal inhibitory neurotransmitter, by allowing chloride ions to enter the neuron upon GABA binding, thereby hyperpolarizing the cell and reducing its activity[2][1][3][4]. GABA_A receptors are composed of a large repertoire of subunits (α, β, γ, δ, ρ, etc.), resulting in substantial functional and pharmacological diversity[2][4]. The receptor distinguishes itself pharmacologically by possessing numerous allosteric sites targeted by drugs such as benzodiazepines, barbiturates, neurosteroids, general anesthetics, and ethanol, which can modulate its activity[2][1]. Subunit composition dictates tissue distribution, pharmacological response, and developmental and pathological regulation. GABA_A receptors are central in the modulation of anxiety, sleep, seizures, muscle tone, and cognition, and thus are implicated in a wide spectrum of neurological and psychiatric disorders[2][1][3][4].
