Overview

(S)-nicotine is a bicyclic alkaloid found predominantly in tobacco plants (Nicotiana tabacum, Nicotiana rustica), consisting of a pyridine ring linked to a pyrrolidine moiety with S-configuration at its chiral center[1][2][6][7]. This enantiomer is the most potent and biologically active form, absorbed via skin, lungs, or digestive tract, and acts as an agonist of nicotinic acetylcholine receptors in the nervous system[4][3][6]. Pharmacologically, it is highly addictive and is both a substance of abuse and a component of smoking cessation therapies[5][3]. Cotinine, a primary nicotine metabolite, is widely used as a biomarker to monitor exposure. Acute toxicity, addiction, and long-term cardiovascular and neurological effects are significant therapeutic challenges[5].
