1,3-1,4-beta-D-glucan, commonly known as mixed-linkage glucan (MLG), is a linear polysaccharide composed of D-glucopyranosyl units linked by a combination of beta-(1->3) and beta-(1->4) glycosidic bonds (PubChem CID 439263). It is a major structural component of the cell walls in cereal grains such as oats and barley, and is also produced by certain bacteria and lichens (Journal of Cereal Science, 2010). While it is not a therapeutic target protein like a receptor or enzyme, it is a bioactive dietary fiber with significant clinical relevance in metabolic health (FDA 21 CFR 101.81). Its primary mechanism involves the formation of a high-viscosity gel in the small intestine, which traps bile acids and slows the absorption of glucose, thereby lowering LDL cholesterol and improving glycemic control (Nutrients, 2019). Additionally, it can act as an immunomodulator by interacting with pattern recognition receptors such as Dectin-1 (CLEC7A) on immune cells, although its activity is distinct from the more common fungal beta-1,3-glucans (Frontiers in Immunology, 2020). Clinically, it is utilized as a nutritional intervention for cardiovascular disease and type 2 diabetes rather than being a target for drug inhibition (EFSA Journal, 2011).
