FKS1 is the catalytic subunit of the 1,3-beta-D-glucan synthase complex, an integral membrane protein essential for synthesizing β-1,3-glucan—a major structural polysaccharide in fungal cell walls. This enzyme uses UDP-glucose as a substrate to catalyze the formation of β(1→3) glycosidic linkages in glucans. It is a key target for echinocandin-class antifungal drugs like caspofungin. Mutations within FKS1, such as S643P, confer resistance to these drugs.
