14-3-3 proteins are a highly conserved family of regulatory molecules found in all eukaryotic cells. They function primarily as scaffolding or adaptor proteins, facilitating protein-protein interactions that are essential for numerous cellular processes, including metabolism, signal transduction, cell cycle control, apoptosis, protein trafficking, transcription regulation, stress responses, and malignant transformation.
14-3-3 proteins bind to target proteins containing phosphorylated serine/threonine residues within specific sequence motifs. This interaction modulates the activity or localization of their binding partners by inducing conformational changes, masking or exposing functional domains, and/or facilitating complex formation with other signaling molecules.
