15-hydroxyprostaglandin dehydrogenase (15-PGDH), encoded by the HPGD gene, is a key enzyme responsible for the metabolic inactivation of prostaglandins. It catalyzes the NAD+-dependent oxidation of the 15-hydroxyl group of prostaglandins to yield inactive 15-keto metabolites. Loss or downregulation of HPGD expression has been observed in several cancers, leading to increased levels of pro-tumorigenic prostaglandins. It acts as an anti-inflammatory mediator and a tumor suppressor.
