17β-Hydroxysteroid dehydrogenase type 13 (HSD17B13) is an enzyme primarily expressed in the liver and associated with lipid droplets. It belongs to the short-chain dehydrogenase/reductase (SDR) family and catalyzes oxidation-reduction reactions involving lipid substrates like retinol. While initially thought to be involved in steroid metabolism, it primarily contributes to hepatic fatty acid metabolism. HSD17B13 is upregulated in NAFLD, and loss-of-function mutations are protective against cirrhosis and HCC. It interacts with ATGL, influencing lipolysis and lipid accumulation. Therapeutic strategies focus on inhibition or modulation of its activity to treat NAFLD/NASH.
