Overview

The AAA+ ATPase subunits (Rpt1-Rpt6 or PSMC1-PSMC6) of the 19S regulatory particle are essential components of the eukaryotic ubiquitin-proteasome system (UPS). They form a heterohexameric ring that uses ATP hydrolysis to unfold target proteins and translocate them into the 20S core particle for degradation. The C-terminal tails of these ATPases are critical for opening the gate of the 20S particle. Dysregulation or mutation can contribute to diseases like cancer, making them potential therapeutic targets.
