Gankyrin is a small, structurally-repeat protein consisting of seven ankyrin repeats that functions as a non-ATPase subunit of the 19S regulatory cap of the 26S proteasome. It plays critical roles in cell cycle progression and oncogenesis by promoting the degradation of key tumor suppressors (p53 and pRb) and by affecting the activity of CDK4 and MDM2, contributing to cellular transformation and proliferation. Gankyrin interacts with several other proteins through its conserved ankyrin repeat surface, acting as a hub for protein-protein interactions critical for both proteasome assembly and cancer cell signaling. Its overexpression is highly correlated with cancer progression and poor prognosis, making it an attractive target for anti-cancer drug development and a clinical biomarker for aggressive tumors.
Inhibitors may block gankyrin’s interactions with CDK4, MDM2, and proteasome machinery, which in turn can stabilize tumor suppressors p53 and pRb
