Overview

26S proteasome non-ATPase regulatory subunit 11 (PSMD11) is a non-ATPase subunit of the 19S regulatory "lid" of the 26S proteasome complex, a multiprotein machinery responsible for ATP-dependent degradation of ubiquitinated proteins in eukaryotic cells. PSMD11 is essential for proteasome assembly and overall proteolytic activity, impacting processes such as cell cycle progression, apoptosis, DNA repair, antigen presentation, and regulation of circadian clock activity via control of negative regulators' stability and nuclear translocation. Aberrant PSMD11 expression has been associated with tumorigenesis and may influence immune and cancer cell biology, making it a candidate diagnostic/prognostic and therapeutic target, particularly in oncology.
