Overview

The 26S proteasome regulatory subunit 6B (PSMC4) is an essential ATPase component of the 19S regulatory particle of the 26S proteasome [UniProt: P43686]. It belongs to the AAA+ (ATPases Associated with diverse cellular Activities) family and forms part of the heterohexameric ring that powers the unfolding and translocation of ubiquitinated proteins into the 20S proteolytic core [PubMed: 25470037]. PSMC4 is critical for maintaining cellular protein homeostasis, regulating the cell cycle, and modulating signal transduction pathways [PubMed: 16330667]. In clinical practice, the 26S proteasome is a major therapeutic target for hematological malignancies like multiple myeloma, where inhibitors such as Bortezomib disrupt the degradation of regulatory proteins to induce apoptosis [PubMed: 28154168]. Beyond cancer, PSMC4 is implicated in neurodegenerative diseases due to its role in clearing misfolded protein aggregates [PubMed: 30210135]. Emerging research also explores PSMC4 as a potential biomarker for treatment response and a target for novel proteasome-modulating therapies [PubMed: 33005034].
