Overview

28S ribosomal protein S21, mitochondrial (MRPS21) is a nuclear-encoded protein that forms part of the small 28S subunit of the mammalian mitochondrial ribosome (mitoribosome). MRPS21 assists in mitochondrial translation, enabling the synthesis of 13 essential proteins encoded by mitochondrial DNA that are required for oxidative phosphorylation and cellular energy production. Mitochondrial ribosomes (28S small + 39S large subunits) differ in composition from prokaryotic ribosomes and are characterized by a higher protein/rRNA ratio and unique sequence and structural features that are critical for mitochondrial-specific translational needs. Mutations or reduced expression of MRPS21 and other mitochondrial ribosomal proteins can result in mitochondrial dysfunction and have been associated with inherited human disorders such as combined oxidative phosphorylation deficiency[1][2][3][6]. There is currently no evidence that MRPS21 is a direct drug target nor associated with specific pharmacological agents.
