3β-Hydroxysteroid dehydrogenase type I (3β-HSD1) is a key enzyme in the biosynthesis of all classes of active steroid hormones. It catalyzes the oxidation and isomerization of Δ5-3β-hydroxysteroids to Δ4-3-ketosteroids, which are essential steps in producing progesterone, glucocorticoids, mineralocorticoids, androgens, and estrogens. HSD3B1 is predominantly expressed in placenta and peripheral tissues such as breast tissue and is also found in some tumors including breast cancer. Deficiency or mutations in HSD3B1 can lead to disorders such as congenital adrenal hyperplasia. Overexpression or altered activity has been implicated in hormone-dependent cancers like breast cancer.
