3-phosphoinositide-dependent protein kinase 1 (PDK1 or PDPK1) is a serine/threonine kinase that acts as a central regulator in multiple signaling pathways, particularly those downstream of phosphatidylinositol 3-kinase (PI3K). It is a master kinase for members of the AGC family of kinases, phosphorylating and activating them at their activation loop, which is essential for their full activation. Through its targets, PDK1 regulates critical cellular processes such as cell survival, growth, proliferation, metabolism, differentiation, and translation. It is involved in cancer and other diseases.
