Overview

39S ribosomal protein L33, mitochondrial (MRPL33) is a nuclear-encoded structural protein forming part of the large (39S) subunit of the mitochondrial ribosome in humans. It is responsible for aiding mitochondrial protein synthesis, a critical process for the function of mitochondria. Mitochondrial ribosomal proteins like MRPL33 differ significantly in sequence and biochemical properties from their prokaryotic counterparts, making sequence-based identification challenging. Dysfunction of MRPL33 has been associated with mitochondrial diseases such as Leigh Disease and may have a role in mitochondrial-related tumor progression[1][2][7][8][9]. MRPL33 is not a classical drug target such as a receptor, enzyme, or transporter, but a ribosomal structural protein. There is no evidence it is directly targeted by approved drugs or used as a biomarker in clinical practice. Alternative splice variants exist; MRPL33's main function is as part of the mitochondrial translation machinery[1][2][6][7].
