Overview

40S ribosomal protein S14 is a cytoplasmic structural component of the eukaryotic small ribosomal subunit (40S), belonging to the S11P family of ribosomal proteins. It is essential for protein synthesis and ribosome stability. While not a receptor, enzyme, or transporter, RPS14 participates in fundamental biological processes, and its mutation or deficiency is associated with bone marrow failure syndromes and cancer risk. Certain drugs like emetine interact with the ribosome, and resistance may arise from mutations in RPS14. Loss of RPS14 is specifically implicated in the clinical features of 5q- syndrome, including anemia and risk of acute myeloid leukemia. Because ribosomal proteins are vital for survival, they are generally not considered therapeutic drug targets but may serve as biomarkers in hematological disorders.
