Small ribosomal subunit protein uS19 is a highly conserved eukaryotic ribosomal protein that is a core component of the 40S small ribosomal subunit[2][5]. Also known as 40S ribosomal protein S15, uS19 is involved in several essential cellular functions. Its conserved globular domain helps form the head of the small ribosomal subunit, while a eukaryote-specific C-terminal extension directly participates in constructing the decoding site, impacting translational fidelity in a manner unique to eukaryotes[2]. uS19 is crucial for 40S ribosome maturation, interacting with assembly factors needed for nuclear export of pre-40S particles. Beyond ribosome biogenesis and translation, uS19 also has extra-ribosomal roles, most notably through regulation of the tumor suppressor p53 in the Mdm2/Mdmx-p53 pathway[2]. Mutations in the RPS15 gene encoding uS19 have been linked to Diamond Blackfan anemia, chronic lymphocytic leukemia, and Parkinson's disease, likely due to defects in ribosome biogenesis or altered translational fidelity[2]. Note: There are no established drugs specifically targeting uS19, nor is it generally considered a "therapeutic target" such as a receptor, enzyme, or transporter. It is better classified as a core structural and functional component of the ribosome[2][5]. No known biomarkers, mechanisms of drug action, or specific safety concerns are associated with antagonizing or modulating uS19 directly, as would be the case with classical drug targets.
