Overview

40S ribosomal protein S28 (RPS28) is a cytoplasmic protein component of the small (40S) ribosomal subunit and belongs to the S28E family of ribosomal proteins. As part of the ribosome, RPS28 is essential for protein synthesis and ribosome biogenesis, contributing to the assembly and structural integrity of the small subunit. It also plays a regulatory role in the processing of pre-rRNA and modulates peptide generation for MHC class I immunosurveillance by influencing the translation initiation from non-canonical start codons. Mutations in RPS28 can result in bone marrow failure syndromes, particularly Diamond-Blackfan anemia with craniofacial abnormalities, and broader ribosomopathies, highlighting its importance in hematopoiesis and development. No direct drugs target RPS28, and it is not considered a typical therapeutic target, but its dysfunction has diagnostic and clinical relevance in specific anemias and developmental disorders.
