Overview

40S ribosomal protein S30 (FAU) is a human protein encoded by the FAU gene, which produces a fusion protein consisting of an N-terminal ubiquitin-like domain (FUBI) and a C-terminal ribosomal protein S30[1][2][3][4]. In cells, the fusion is cleaved post-translationally, separating FUBI from S30; only S30 becomes part of the mature 40S ribosomal subunit and acts in the cytoplasmic ribosome to facilitate mRNA translation[1][2][4]. FUBI shares moderate structural similarity with ubiquitin but its cellular roles are less defined; however, both intact FAU and its FUBI domain have been implicated in the regulation of apoptosis (programmed cell death) through interactions with proteins such as Bcl-G, and the gene is considered a potential tumor suppressor[2]. Downregulation of FAU may increase resistance to apoptosis and be linked to tumor progression in cancer, suggesting its function as a biomarker for tumor suppression and prognosis[2]. There are currently no well-established therapeutics or drugs known to specifically target FAU directly, and drug mechanism-of-action or safety profiles are not documented for this protein.
