Overview

40S ribosomal protein S9 (RPS9) is a component of the small (40S) subunit of the eukaryotic ribosome. It plays a crucial role in ribosome biogenesis, directly binds ribosomal RNA during early ribosome assembly, and is essential for the translation of mRNA into proteins within the cytoplasm. RPS9 is part of the S4P family of ribosomal proteins and is highly conserved across eukaryotes, prokaryotes (as RPS4), and archaea. In human cells, RPS9 is detected in both the nucleolus (site of ribosome assembly) and the cytoplasm (site of active translation). Mutations or depletion of RPS9 disrupt ribosome production, impair cell proliferation, and can induce cell cycle arrest or apoptosis. Variable expression has been observed in certain cancers, such as colorectal cancer, and it has mechanistic roles in diseases like Diamond-Blackfan anemia and Takayasu arteritis
