Overview

The 5’-deiodinase enzymes (D1, D2, and D3) are a family of selenoproteins that regulate thyroid hormone activity by catalyzing the removal of iodine atoms from thyroid hormones. D1 and D2 activate T4 to T3, while D3 inactivates T4 and T3. This regulation is crucial for maintaining tissue-specific levels of active thyroid hormone and for developmental control. Alterations in these enzymes can disrupt thyroid hormone homeostasis, leading to disorders such as hypothyroidism/hyperthyroidism or abnormal neurodevelopment.
