Overview

5α-reductase type 2 is an enzyme that catalyzes the conversion of testosterone to dihydrotestosterone (DHT), a more potent androgen essential for male sexual differentiation. It is encoded by the *SRD5A2* gene. Deficiency or mutations in *SRD5A2* cause disorders of sex development (DSDs) such as ambiguous genitalia or undervirilization in individuals with XY chromosomes. In prostate cancer, the expression of type 2 isoform decreases while type 1 increases. Antiandrogen therapies can block this regulatory mechanism.
