The 5S ribosomal RNA (5S rRNA) is a small, highly conserved, non-coding RNA (approximately 120 nucleotides) that is an essential structural and functional component of the large ribosomal subunit in all domains of life apart from fungal and animal mitochondria. In eukaryotes, multiple copies of the 5S rRNA gene reside in tandem repeats. 5S rRNA participates in the assembly of the ribosome and mediates interactions among key functional centers of the ribosome, indirectly influencing protein synthesis rates and, in some contexts, p53 regulatory activity. The 5S rRNA gene “5S rRNA 5” or “RNA5S5” refers to a single member of the tandemly repeated gene family and is not a unique molecular drug target. It is not considered a classic therapeutic target, as it is not a protein, receptor, enzyme, or transporter, but rather a core ribosomal RNA required for cell viability and ribosome assembly.
