Overview

5S ribosomal RNA (5S rRNA) is a small (~120 nucleotides) non-coding RNA that is an essential structural and functional component of the large subunit of the ribosome in all domains of life except the mitochondria of animals and fungi[1][2][3]. It participates in the assembly and structural integrity of the ribosome, bridging the peptidyl transferase center and the GTPase-associated center and helping communicate between key domains during protein synthesis[1][2][3]. The “8” designation appears to reference a specific gene or isoform, but “5S ribosomal RNA 8” is not a standard, widely recognized nomenclature; in human genome annotation, multiple 5S rRNA genes exist, often labeled by genomic locus rather than function. This RNA is not a receptor, enzyme, transporter, or classical therapeutic target, nor is it individually targeted by drugs[2][3]. While dysfunction of ribosomal RNA can contribute to diseases associated with ribosome biogenesis (ribosomopathies), 5S rRNA isoforms themselves are not direct disease targets or commonly used biomarkers[1][2][3]. The name “RNA5S8” is non-standard and does not correspond to an established pharmacological or biological target.
