Overview

5T4, also known as trophoblast glycoprotein (TPBG), is a 72 kDa heavily N-glycosylated transmembrane protein and a member of the leucine-rich repeat (LRR) family. It is highly expressed in placental trophoblasts and a wide range of carcinomas, but has limited expression in most normal adult tissues. 5T4 promotes EMT, potentiates chemotaxis signaling, and modulates Wnt pathways, contributing to tumor initiation, progression, invasion, metastasis, and recurrence. High levels of 5T4 are associated with poor prognosis in multiple cancers, making it a therapeutic target for cancer vaccines, antibody-drug conjugates, tumor-targeted superantigens, and CAR-T cell therapies.
