Overview

60S ribosomal export protein NMD3 (NMD3) is a highly conserved, essential adaptor protein required for the export of the 60S ribosomal subunit from the nucleus to the cytoplasm in eukaryotic cells[1][2][3][4]. NMD3 binds to newly assembled 60S ribosomal subunits in the nucleus, where it provides a leucine-rich nuclear export signal (NES) recognized by the export receptor CRM1 (Exportin 1), enabling the complex to transit the nuclear pore complex in a RanGTP-dependent manner[2][3]. NMD3 is released from the subunit after export, in a process dependent on the ribosomal protein Rpl10 and the GTPase Lsg1, thus permitting final maturation and entry of the 60S subunit into the translation-competent pool[1][3][4]. Structurally, NMD3 contains zinc-binding centers and mimics certain translation factors during ribosome biogenesis[4]. NMD3's conserved role across eukaryotes and its function in ensuring only mature, functional 60S subunits are exported underscore its centrality in cellular protein synthesis and growth[1][2][3][4]. There are no currently approved drugs acting directly on NMD3, and its indispensable role in ribosome production means it is not considered a typical therapeutic target.
