60S ribosomal protein L13 (RPL13) is a highly conserved protein that is a component of the large (60S) ribosomal subunit, essential for the synthesis of cellular proteins[1][2][3]. RPL13 is part of the L13E family of ribosomal proteins and is located in the cytoplasm. Besides its canonical role in protein synthesis, RPL13 has emerging extraribosomal functions: it regulates cell proliferation, participates in cellular stress responses, and plays critical roles in bone and heart development[3]. It is upregulated in various cancers and is associated with disease progression and chemoresistance. RPL13 also participates in the innate immune response and antiviral defense mechanisms, modulating NF-κB and interferon signaling pathways[4]. Mutations in RPL13 cause certain rare congenital skeletal disorders. There are no approved drugs that directly target RPL13, and it is not currently considered a direct therapeutic target.
