60S ribosomal protein L22 (RPL22) is a structural protein component of the large (60S) subunit of the eukaryotic ribosome and is integral to protein synthesis in the cytoplasm[3][5]. It is part of the L22E family, interacts directly with ribosomal RNA (notably 23S/28S rRNA), and has the ability to bind certain viral RNAs such as those from Epstein-Barr virus (EBV)[3]. In mice, RPL22 is essential for normal T and B cell development and can regulate its own function as well as that of the closely related RPL22-like1 (RPL22L1), which can compensate for its absence to some extent[1]. Aside from its core structural role, RPL22 may participate in the regulation of RNA stability, telomerase activity, and be involved in extra-ribosomal activities relevant to immune function and disease development[1][3]. There is no evidence that RPL22 functions as a classical drug target, receptor, enzyme, or transporter; its medical significance derives from its indirect roles in disease pathogenesis.
