Overview

60S ribosomal protein L28 (RPL28) is a structural protein component of the large (60S) subunit of the eukaryotic ribosome, belonging to the L28E family of ribosomal proteins[1][2][3][4]. Ribosomes are essential organelles responsible for translating messenger RNA (mRNA) into proteins in all living cells. RPL28 is located in the cytoplasm and participates directly in the assembly and stability of the ribosome by interacting with ribosomal RNA[1][2][3][4]. Expression of RPL28 is ubiquitous, but variable expression has been observed in colorectal cancer tissue compared to adjacent normal tissue; elevated tumor RPL28 levels and a particular germline SNP (rs4806668G>T) have been linked to poorer survival in metastatic colorectal cancer, making RPL28 a candidate prognostic biomarker and an indirect potential target for cancer research, though it is not typically considered a direct drug target[3][4]. Disruption of RPL28 expression in cancer cell lines reduces cellular proliferation, suggesting a functional contribution to tumor biology[3]. Diseases linked to ribosomal dysfunction such as Diamond-Blackfan anemia and certain motor neuron diseases have also been associated with RPL28, although data are limited[4]. No drugs are known to directly target this protein, and due to its essential role in protein synthesis, non-specific inhibition would likely have broad cytotoxic effects.
