Overview

60S ribosomal protein L29 (RPL29) is a cytoplasmic ribosomal protein and a structural component of the 60S large ribosomal subunit in eukaryotic cells[1][6]. It is essential for proper ribosome biogenesis and global protein synthesis. In addition to its primary location in the ribosome, RPL29 is also expressed as a peripheral membrane protein on the cell surface, where it binds heparin and heparan sulfate, a property linked to modulation of angiogenesis and interaction with growth factors[1][2][5]. Functionally, RPL29 has roles in regulating cell proliferation, differentiation, and apoptosis—knockdown studies implicate it in cancer cell growth arrest and apoptosis, as well as the modulation of immune cell cytotoxicity[3]. Altered expression of RPL29 has been reported in various cancers and is associated with tumor cell behavior and tumorigenesis[3]. There are currently no approved drugs known to selectively target RPL29, and it is not a therapeutic target at this time; its essential role in ribosomal function makes it a potential risk for off-target effects if inhibited[3][6].
